ABSTRACT

ObjectiveTo study the effect of Bazi Bushen capsules on delaying the aging process of naturally aging mice and its mechanism. MethodThe mice were randomly divided into four groups according to their body weight, namely, aging group, low-dose Bazi Bushen capsules group (1 g·kg-1), high-dose Bazi Bushen capsules group (2 g·kg-1), and rapamycin group (0.002 g·kg-1). The debilitating signs were detected by behavioral tests and the weakness index was measured. The percentages of spleen T and B lymphocytes, effector T cells (TE), memory T cells (TM), naive T cells (TN), helper T cells (Th), cytotoxic T cells (Tc) ,Th1 cells, Th2 cells, and regulatory T cells (Treg) were determined by flow cytometry. Cell proliferation and the cell counting kit-8 (CCK-8) assay was used to detect the proliferation of lymphocytes in mice. The hematoxylin-eosin (HE) staining was used to observe the histopathological changes in the mouse spleen. The expression of cyclin-dependent kinase inhibitor 2A (p16) and cyclin-dependent kinase inhibitor 1A (p21) was detected by immunohistochemistry (IHC). The mRNA expression of senescence-related proteins p16 and p21 was determined by real-time fluorescence quantitative polymerase chain reaction (Real-time PCR), and the inflammatory cytokines, including tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), interleukin (IL)-1β, IL-2, IL-4, IL-10, and IL-12 p70, in peripheral blood of mice were detected by Luminex. ResultAs compared with the aging group, mice in the Bazi Bushen capsules and rapamycin groups showed significantly improved debilitating signs and reduced weakness index scores (P<0.05, P<0.01), increased proportions of T cells, TN cells, Tc cells, Th2 cells, and Treg cells in the spleen, decreased proportions of TE cells, TM cells, Th cells, Th1 cells (P<0.05, P<0.01), and increased proliferation of splenic lymphocytes (P<0.05, P<0.01). In the Bazi Bushen capsules and rapamycin groups, clear structure of the red and white marrow marginal zone was observed in the spleen of mice, the area of the white marrow was increased, and the area of the red marrow was correspondingly decreased. The protein and mRNA expression of aging-related proteins p16 and p21 in the spleen was decreased (P<0.01), the levels of serum pro-inflammatory cytokines IL-1β, IL-2, IL-12 p70, IFN-γ, and TNF-α levels were decreased, and the levels of IL-4 and IL-10 were increased (P<0.05, P<0.01) in the Bazi Bushen capsules and rapamycin groups as compared with the aging group. ConclusionBazi Bushen capsules have the effect of regulating the debilitating signs of natural aging mice, regulating the immune homeostasis and inflammation level of the body, and reducing cell aging.

ABSTRACT

Objective:To screen the serum exosomal microRNAs differentially expressed in early pancreatic cancer patients and evaluate the diagnostic value of exosomal hsa-let-7f-5p.Methods:From January 2019 to January 2020, 19 patients with early pancreatic cancer (early pancreatic cancer group) and 16 patients with chronic mass-forming pancreatitis (pancreatitis group) were selected from Affiliated Hospital of Nanjing University of Chinese Medicine who underwent surgery and were confirmed by pathology. Serum samples of the two groups of patients were collected. At the same time, serum samples of 19 healthy volunteers were selected as the normal control group. The exoEasy Maxi Kit was used to isolate serum exosomes. The structural characteristics of exosomes were observed by transmission electron microscopy (TEM). The particle size of exosomes was observd by nanoparticle tracking analysis. CD 63 and CD 81, the specific protein marker on the surface of exosomes, were identified by western blotting. The total RNA of exosomes was extracted by the miRNeasy Serum/Plasma Kit, and a small RNA library was constructed after quality inspection. With reference to the small RNA database, the differentially expressed exosomal microRNAs in early pancreatic cancer group, pancreatitis group and normal control group were filtered out. The miRNA candidates were validated by quantitative polymerase chain reaction (qPCR) and different expressions of them were analyzed. The role of target genes and metabolic pathways of candidate miRNAs in the occurrence and development of early pancreatic cancer were analyzed by gene ontology (GO) and Kyoto Encyclopeda of Genes and Genomes(KEGG) enrichment pathway. Results:The isolated serum exosomes can be seen to have cup-like vesicle with the double lipid layer by TEM. The main peak of the particle size of target exosomes was about 150 nm. The expression of exosome specific protein markers CD 63 and CD 81 was positive. Comparing the expression of miRNAs among early pancreatic cancer group, pancreatitis group and normal control group, the specific tumor marker exosomal hsa-let-7f-5p was screened out in this study, and its expression in early pancreatic cancer group was significantly higher than that in pancreatitis group and normal control group (both P values <0.05). Receiver operating characteristic curve analysis (ROC) showed that the area under curve (AUC) of exosomal hsa-let-7f-5p to distinguish pancreatic cancer from pancreatitis was 0.843 (95% CI 0.640-1.000). The sensitivity and specificity were 100% and 81.82% respectively. The AUC for distinguishing pancreatic cancer from normal controls was 1.000 (95% CI 1.000-1.000), and both sensitivity and specificity were 100%. The diagnostic efficiency of exosomal hsa-let-7f-5p was equivalent to that of CA19-9 ( P>0.05). The GO analysis results showed that target genes of exosomal hsa-let-7f-5p were mainly involved in complement activation lectin pathway in biological processes, and the proteins expressed by target genes were mainly distributed in cilium, and molecules mostly functioned by combining with nitric-oxide synthase. The KEGG pathway enrichment analysis showed that the target genes were closely related to MAPK signaling pathway. Conclusions:Serum exosomal hsa-let-7f-5p has the potential to be a diagnostic biomarker for early pancreatic cancer.

ABSTRACT

Objective To analyze the prevalence of myopic refractive error and its associated risk factors among primary school students in Shanghai Minhang Distict. Methods A total of 11 356 primary school students who participated in the children's refraction screening project in Meilong Community, Minhang District, Shanghai from January 1 to December 31, 2019 were selected for visual inspection and computer optometry Results The detection rate of myopic refractive error among 6-10 year-old students was 45.59%, with a higher rate in girls and older age group. The detection rates of 6-10 year-old students were 22.88% (6 year-old), 31.77% (7 year-old), 47.62% (8 year-old), 60.53% (9 year-old) and 69.84% (10 year-old), respectively, showing a linear trend(χ 2=1.31, P<0.001). Multivariate logistic regression revealed that myopic refractive error was statistically significantly associated with gender, age and parents' myopic. Birth weight, preterm birth and body mass index (BMI) had no influence on refractive myopic in primary school students. Conclusion Myopic refractive error is still a major public health problem in primary school students. A combined strategies for myopic refractive error reduction should be called to focus on high risk persons and behaviors.

ABSTRACT

Hashimoto’s thyroiditis (HT) is one of the most common autoimmune diseases, and often combined with papillary thyroid carcinoma (PTC) .Thyroglobulin antibody and thyroid peroxidase antibody are used as HT marker antibodies and effective indicators for diagnosis. By activating the corresponding immune response, chronic inflammation of the thyroid gland is caused, which is related to the development of tumors. In this review, we analyze the significance of Tg and TgAb in diagnosis and treatment of HT-PTC to provide evidence for future clinical studies.

ABSTRACT

At present, due to the opening and circulation of salt market, potassium iodate iodized salt, potassium iodide iodized salt, seaweed iodized salt and non-iodized salt may exist simultaneously in the edible salt market of a region, which brings new problems to the work of testing iodine content of edible salt in the laboratory. In order to carry out salt iodine detection for prevention and monitoring of iodine deficiency disorders, this paper introduces some methods which established by the author for a rapid qualitative, semi-quantitative and quantitative detection of iodized salt of different forms of iodine and non-iodized salt. And further application and discussion of the methods are described for reference of iodized salt monitoring.

ABSTRACT

Development of thoracolumbar vertebra (TLV) and rib primordium (RP) is a common evolutionary feature across vertebrates, although whole-organism analysis of the expression dynamics of TLV- and RP-related genes has been lacking. Here, we investigated the single-cell transcriptome landscape of thoracic vertebra (TV), lumbar vertebra (LV), and RP cells from a pig embryo at 27 days post-fertilization (dpf) and identified six cell types with distinct gene expression signatures. In-depth dissection of the gene expression dynamics and RNA velocity revealed a coupled process of osteogenesis and angiogenesis during TLV and RP development. Further analysis of cell type-specific and strand-specific expression uncovered the extremely high level of HOXA10 3'-UTR sequence specific to osteoblasts of LV cells, which may function as anti-HOXA10-antisense by counteracting the HOXA10-antisense effect to determine TLV transition. Thus, this work provides a valuable resource for understanding embryonic osteogenesis and angiogenesis underlying vertebrate TLV and RP development at the cell type-specific resolution, which serves as a comprehensive view on the transcriptional profile of animal embryo development.

ABSTRACT

Objective:To explore the of short-term psychodynamic psychotherapy on anxiety and depression of gout patients and its influence on well-being index, family function and compliance.Methods:60 gout patients were selected from June 2018 to December 2019 in the outpatient department of rheumatism and immunity department of Dongguan Kanghua Hospital by prospective case-control study. They were randomly divided into control group and treatment group, 30 cases respectively. Both groups were given gout related clinical treatment and routine nursing, and the treatment group was given short-term dynamic orientation psychotherapy. The treatment compliance, the psychological status, well-being index and blood uric acid (UA) level of the two groups were compared before intervention, immediately after intervention and 3 months after intervention.Results:The scores of Patient Health Questionnaire-9 (PHQ-9) and the generalized anxiety disorder 7-item scale (GAD-7) in the two groups were gradually decreased immediately and 3 months after intervention ( P<0.05), and the scores of well-being index were gradually increased ( P<0.05); Compared with the control group, the scores of PHQ-9 and GAD-7 in the treatment group decreased more significantly ( P<0.05), and the score of well-being index increased more significantly ( P<0.05); The scores of communication, behavior control and total function of family function in the treatment group decreased gradually immediately after intervention and 3 months after intervention ( P<0.05), and were significantly lower than those in the control group ( P<0.05); The level of serum UA in the two groups decreased gradually immediately after intervention and 3 months after intervention ( P<0.05), but there was no significant difference in UA level at each time point between the two groups. The treatment compliance of the treatment group was significantly better than that of the control group ( P<0.05). Conclusions:The short-term dynamic orientation psychotherapy for gout patients has obvious effect in improving the state of anxiety and depression, improving the index of well-being, family function and treatment compliance.

ABSTRACT

Objective:To develop a standard method for determination of iodine in serum by inductively coupled plasma mass spectrometry (ICP-MS).Methods:A direct dilution sampling-ICP-MS method for measuring iodine in serum was developed with 2.0 g/L ascorbic acid-1.0 g/L ammonium chloride-0.10% ethanolamine-1.0% ethanol as diluent; the standard solutions and the serum samples were all diluted in a ratio of 1 : 19 (sample : diluent) before testing with Re as internal standard element. Methodological evaluation of a new method was done through standard curve linearity, sample detection limit, precision and accuracy in determining serum iodine. And the results were compared with the results determined by current standard method for determination of iodine in serum [arsenic-cerium catalytic spectrophotometry, hereinafter referred to as the standard method (WS/T 572-2017)].Results:The following data were obtained by testing the method in six laboratories. The linear range of the calibration curve was 0 - 300 μg/L and the linear correlative coefficients were 0.999 9 - 1.000 0 ( n = 70). The detection limit for serum iodine was 0.2 to 1.3 μg/L (0.20 ml of serum was tested). Precision: the average intra assay coefficient of variation ( CV) was 0.6% with a range of 0.2% - 1.6% when measuring 31 serum samples; the average inter assay CV was 1.3% with a range of 0.3% - 2.4% when measuring 34 serum samples. Accuracy: the average recovery was 100.3% with a range of 93.9% - 105.6% when measuring 36 serum samples. No significant difference was found between the results of the 116 serum samples determined by the new method and the standard method ( P > 0.05). Conclusions:The new method has a good standard curve linearity, high sensitivity, good precision, accuracy and is easy to be used and quickly to be analyzed of the test results, and is suitable for widely application in determining serum iodine.

ABSTRACT

Objective:To investigate the iodine nutritional status among children aged from 8 - 10 and pregnant women in Xiamen City before and after adjustment of salt iodine concentration, and scientifically evaluate the suitability of the new standard iodized salt adjustment policy for Xiamen City.Methods:In the pre-adjustment period (2011) and the post-adjustment period (2018), cross-sectional research methods and stratified cluster sampling methods were adopted in six districts of Xiamen City. The salt samples of residents and pregnant women, urine samples of children aged from 8 - 10 and pregnant women were collected to determine the iodine level; thyroid of children aged from 8 - 10 was examined. Changes in salt iodine, urine iodine and goiter prevalence before and after adjusting iodine content of salt were compared.Results:The median of salt iodine in residents after the adjustment (23.5 mg/kg) was significantly lower than that of before (28.9 mg/kg, Z = - 10.512, P < 0.05), the median of salt iodine in pregnant women after the adjustment (23.7 mg/kg) was significantly lower than that of before (29.2 mg/kg, Z = - 12.622, P < 0.05); the consumption rate of qualified iodized salt in residents after the adjustment (95.7%, 1 196/1 250) was significantly lower than that of before (97.1%, 1 608/1 656; χ 2 = 4.250, P < 0.05); before and after adjustment of salt iodine concentration, the consumption rate of qualified iodized salt in pregnant women was 94.7% (124/131) and 95.9% (579/604), respectively. No significant difference was found when comparing the consumption rate of qualified iodized salt in pregnant women (χ 2 = 0.015, P > 0.05). There was significant difference in median urinary iodine (from 204.2 to 183.9 μg/L, detected in 652, 1 250 urine samples, respectively) of children aged 8 - 10 years ( Z = - 3.583, P < 0.05); the difference of iodine nutritional status in pregnant women before and after adjustment of salt iodine concentration(130.2, 130.8 μg/L, detected in 132, 604 urine samples, respectively) was not significant ( Z = - 1.715, P > 0.05). The thyroid goiter rate of children aged 8 - 10 years had reduced from 1.1% (14/1 239) to 0.2% (2/1 253), the difference was statistically significant (χ 2 = 9.195, P < 0.05). Conclusions:As a whole, the measure of control and prevention of iodine deficiency disorders through iodized salt is implemented well in Xiamen City. After adjustment of salt iodine concentration, the iodine status of children aged from 8 to 10 is adequate, but the pregnant women has showed iodine deficiency.

ABSTRACT

Objective:To build a medical service experience system based on role play of hospital staff, and explore its contribution to upgrading medical service, elevating medical service quality, and improving patients′ satisfaction.Methods:An indicator system for medical service experience was developed via literature review. 20 newly recruited hospital employees in 2019 were sampled randomly to form a service experience team, who were arranged to experience the full medical service process of outpatients and inpatients based on role play. The questionnaire for service quality based on patient perception and expectation was used to survey the expectation and perception values of those role players on their medical service experiences, and summarize the defects found on such six dimensions as hospital reliability, responsiveness, tangibility, assurance, empathy and cost-effectiveness. After due corrections of these defects, further experiences and patient satisfaction survey were made to assess the effects of such actions taken. Paired t test was used respectively in statistical analysis of service experience value and actual perception value, as well as the perception values before and after the actions taken, and χ2 test was used for a statistical analysis of patient satisfaction. Results:Role players experienced respectively at the emergency, outpatient and inpatient departments of the hospital for a week in August 2019. Statistics of their experiences indicated≥0.50 points difference between the mean expectation and perception values on doctor-patient communication, patient help responsiveness, outpatient process, examination report delivery duration, service attitude, patient trust on medical workers, medical environment and patient privacy protection.Following a 3-month reform, role players found improvements in such aspects as hospital reliability, responsiveness, tangibility, assurance, empathy and cost-effectiveness; patient satisfaction improved sizably in December 2019 over January of the same year( P<0.05). Conclusions:The construction and application of the medical service experience system based on role play of hospital staff prove highly useful in upgrading medical service actions, improving medical service quality, and improving patient satisfaction.

ABSTRACT

ObjectiveTo investigate the clinical effect of elbasvir/grazoprevir in the treatment of patients with genotype 1b chronic hepatitis C (CHC). MethodsA total of 99 patients with genotype 1b CHC and compensated cirrhosis who received elbasvir/grazoprevir treatment for 12 weeks and completed treatment and follow-up for 12 weeks after drug withdrawal in Tianjin Third Central Hospital from December 2018 to October 2019 were enrolled. Related clinical data, serological markers, virological indices, and liver stiffness measurement were collected at baseline, at the end of treatment, and at week 12 after drug withdrawal, and virologic response was observed. The Friedman test and Wilcoxon signed rank sum test were used to observe virologic response rate and the changes in liver function and liver stiffness measurement at the end of treatment and at week 12 after drug withdrawal, and the safety of elbasvir/grazoprevir was evaluated. ResultsFor the 99 patients treated with elbasvir/grazoprevir for 12 weeks, the proportion of patients with HCV RNA below the lower limit of detection was 100% at the end of treatment and 99% at week 12 after drug withdrawal. There were significant reductions in alanine aminotransferase (ALT) and aspartate aminotransferase (AST) from baseline to the end of treatment (Z=-5.857 and -5.941, both P<0.05). Liver stiffness measurement decreased from 10.5 kPa at baseline to 8.0 kPa at week 12 after drug withdrawal (Z=-4.036, P<0.05). Among the 99 patients, 24 patients with compensatory cirrhosis reached a virologic response rate of 100% at the end of treatment and at week 12 after drug withdrawal, as well as significant reductions in ALT and AST from baseline (both P<0.05), and liver stiffness measurement decreased from 21.1 kPa at baseline to 17.5 kPa at the end of treatment (Z=-1.832, P=0.067) and 13.6 kPa at week 12 after drug withdrawal (Z=-3.182, P=0.001). Compared with the non-liver cirrhosis group, the liver cirrhosis group had significantly greater reductions in liver stiffness measurement (P<0.05). The patients had good tolerance throughout the treatment, and 4 patients reported mild adverse events during the treatment. ConclusionPatients with genotype 1b CHC have a high virologic response rate to elbasvir/grazoprevir in the real world, with significant improvements in liver function and liver stiffness measurement and good tolerance.

Objective To investigate the correlation between quantitative perfusion histogram parameters of DCE-MRI and tumor tissue microvessel density(MVD) in patients with lung cancer.Methods 30 patients with lung cancer confirmed by pathology who underwent preoperative DCE-MRI were enrolled in this retrospective study .Quantitative perfusion histogram pa-rameters( including median, mean, skewness, kurtosis, energy, entropy) were measured for each patient using Exchange mo-dle.Using the Immunohistochemical method to detect the expression of CD34 in tumor tissue, and counting the number of mi-crovessels under microscope.SPSS 19.0 was used to carry out statistical analysis.The correlation between MVD and quantita-tive perfusion histogram parameters of DCE-MRI measured by exchange model was evaluated by Pearson linear correlation anal-ysis.Results There was no significant difference in MVD and each quantitative perfusion histogram parameters between the three different pathological groups of lung cancer(P >0.05).Ktrans perfusion histogram parameters(mean, 25%, 50%, 50%, 75%, 90%, 95%), Kep perfusion histogram parameters(entropy, 10%, 25%, 50%, 75%, 90%), Fp perfusion histogram parameters( mean, 25%, 50%, 75%, 90%, 95%) and Vp perfusion histograms parameters ( entropy, 75%, 90%, 95% ) were positively correlated with MVD(P<0.05).Ktrans perfusion histogram parameters(energy) and Vp perfu-sion histogram parameters(skewness, kurtosis, energy) were negatively correlated with MVD(P<0.05).There was no signifi-cant correlation between Ve perfusion histogram parameters and MVD(P>0.05).Conclusion There was a certain correla-tion between the perfusion histogram parameters of DCE-MRI and MVD,suggesting that the quantitative perfusion histogram of DCE-MRI in lung cancer can reflect the MVD value of cancer tissue .

Objective To analyze the clinicopathological features and prognosis of immunoglobulin G (IgG)4-related diseases with interstitial nephritis. Methods Forty cases of IgG4-related diseases diagnosed by pathology in our hospital from 2014 to 2018 were collected and their clinicopathological features were analyzed. Four patients with IgG4-related disease with interstitial nephritis were analyzed, including clinical laboratory tests and histopathological features, and immunohistochemical analysis of the type and proportion of renal interstitial infiltrating cells. At the same time, the treatment and prognosis of the patients were analyzed. Results Among 40 cases of IgG4-related diseases, 11 cases had parotid submandibular gland involvement (accounted for 28%), labial gland involvement (7 cases, 18%) and lymph node enlargement (6 cases, 15%). Patients with kidney involvement (4 cases, 10%) all presented with IgG4-related tubulo-interstitial nephritis (IgG4-TIN). All of them were elderly males, and 2 had glomerulopathy. One of them had anti-neutrophil cytoplasmic antibodies (ANCA)-related vasculitis renal damage. The number of CD4-positive cells in renal interstitium was more than CD8-positive cells. Another case complicated with IgA nephropathy. Renal dysfunction occurred in all 4 cases, and serum IgG4 level ranged from 4.65 g/L to 23.8 g/L. All 4 patients received glucocorticosteroid and symptomatic treatment, and the prognosis was good. Conclusion IgG4-related diseases may involve mul-tiple organs, renal dysfunction may occur when the kidney is involved. Interstitial nephritis is the major clinical manifestation. Glomerular lesions may accelerate the progress of IgG4-TIN. Corticosteroid therapy is effective.

Objective To establish a catalytic spectrophotometric method for determination of iodine in water without the use of highly toxic arsenic reagents with good precision and accuracy.Methods A new flow injection catalytic spectrophotometry method for determination of iodine in water was established by combining flow injection analysis (FIA) with a new catalytic spectrophotometry for determination of iodine with no need of arsenic reagent.The standard curve linearity,sample detection limit,precision and accuracy of different iodine content ranges (0-250,0-1 200 μg/L) were tested.The test result was compared with that of the arsenic-based catalytic spectrophotometry recommended by the iodine deficiency disorders monitoring program in China (recommended monitoring method).Results A calibration relation of C =aA2 + bA + d (C:iodine concentration,A:measuring absorbance peak area from FIA) of the new method existed in the range of 0-250 and 0-1 200 μg/L.The linear correlative coefficient were both 0.999 9.The detection limit for iodine in water was 0.3 μg/L.The FIA peak shape of the sample tube with the lowest content is determined,and the sensitive iodine signal is detected.The water samples with different iodine content ranges were determined in parallel for 6 times,and the coefficient of variation (CV) was 0.2%-2.7%;the recoveries ranged from 95.0% to 105.0%,and the total average recovery was 100.3%.The measured values of water iodine standard material GBW 09113e [(8.2 ± 1.2) μg/L] and GBW 09114d [(62.0 ± 6.0) μg/L] in the standard curve of different iodine contents were all within the uncertainty range of the given value.There was no significant difference between the results of recommended monitoring method and the new method (t =0.781,P > 0.05).Conclusions The new method for measuring water iodine has a wide standard curve range,high sensitivity,and good precision and accuracy.The instrument is easy to operate,and no need for toxic arsenic reagent.The method is a reliable method for water iodine detection.

ABSTRACT

Objective To determine serum iodine by inductively coupled plasma mass spectrometry (ICP-MS).Methods ICP-MS was used to determine iodine content directly by pretreatment method of diluting serum samples and standard series with diluent.The method was tested from correlation coefficient of standard curve,detection limit,precision,accuracy,etc.Results When the concentration of iodine in serum ranged from 0 to 300 μg/L,the linear correlation coefficient with the iodine ion counts determined by the instrument was 0.999 8 to 1.000 0 (n =6);the detection limit of iodine in serum was 1.24 μg/L;the relative standard deviation (RSD) ranged from 0.5% to 1.5% and from 0.3% to 1.4% for the intra-and inter-batch precision tests of seven different serum samples,respectively;the recovery rate of 7 serum samples with iodine content 40-230 μg/L ranged from 93.4% to 109.2%,and the total average recovery rate was 102.4%.Conclusions Serum samples need not be pretreated before digestion,and can be directly injected after dilution with diluent without using highly toxic substances.The detection process is automatic,sensitive,precise,accurate and suitable for rapid quantitative determination of serum iodine content.

ABSTRACT

Objective To analyze the iodine nutritional status among pregnant women,breasffeeding women,0-2 years old infants,8-10 years old children,18-45 years old women and men in both urban and rural areas of Xiamen 3 years after promotion of new standard iodized salt.Methods The Huli and Xiang'an were selected as the urban and rural study sites in Xiamen City,from March 2015 to March 2016.Each study site was further divided into five geographic locations (east,south,west,north and center).In each location,one township was randomly selected.In each town,2 tap water samples,60 salt samples of residents,and urine samples of 40 children aged from 8-10 years old,20 pregnant women,12 breastfeeding women,0-2 years old infants,18-45 years old women and men were collected to determine the iodine level.The water iodine and urinary iodine were detected by arsenic cerium catalytic spectrophotometry;salt iodine was detected by direct titration.Results The medians of salt iodine in urban and rural were 23.0 and 25.3 μg/L,respectively.The coverage rates of iodized salt in urban and rural area were 99.3% (298/300) and 94.0% (282/300),respectively.The consumption rates of qualified iodized salt were 93.7% (281/300) and 90.7% (272/300),respectively.The medians of tap water iodine in urban and rural were 4.1 and 4.4 μg/L,respectively.In urban and rural areas,the medians of urinary iodine of pregnant women were 167.6 and 119.6 μg/L,respectively.The medians of urinary iodine of breastfeeding women were 121.6 and 101.2 μg/L,respectively.The medians of urinary iodine of infants were 165.9 and 110.5 μg/L,respectively.The medians of urinary iodine of children were 208.9 and 167.2 μg/L,respectively.The medians of urinary iodine of women aged 18-45 were 132.5 and 154.5 μg/L,respectively.The medians of urinary iodine of men aged 18-45 were 131.7 and 154.5 μg/L,respectively.Conclusions After 3 year promotion of new standard iodized salt,the iodine status of pregnant women in urban,breastfeeding women,0-2 years old infants,children aged 8 to 10 and adults aged 18 to 45 is adequate,but the pregnant women in rural has showed iodine deficiency.So surveillance and health education should be continuously strengthened in the future.Scientific salt iodization should continue to ensure the amount of iodine intake.

ABSTRACT

Objective To screen a suitable reagent system for measuring iodine in serum by direct dilution sampling-inductively coupled plasma mass spectrometry (ICP-MS).Methods Experiments were on sample dilution method for standard solutions and serum samples with six different diluents.After dilution,the concentration of iodine in serum was determined by ICP-MS method.The rhenium (Re) and tellurium (Te) was used as internal standard element,respectively.The effects of the two internal standard elements on determination results were compared.And the results were compared with the results determined by the current serum iodine arsenic cerium catalytic spectrophotometry standard method (WS/T 572-2017),hereinafter referred to as the standard method.The methodological evaluation of this new method was done through standard curve linearity,sample detection limit,precision and accuracy in determining serum iodine in the range of 0-300 μg/L.Results The direct dilution sampling-ICP-MS method screened for measuring iodine in serum was preparing 1 L of 2.0 g/L ascorbic acid,1.0 g/L ammonium chloride,0.10% ethanolamine,and 1.0% ethanol as diluent;the standard solutions and the serum samples were all diluted in a ratio of 1:19 (sample:diluent) before testing with Re as internal standard element.The linear range of the calibration curve was 0-300 μg/L and the linear correlative coefficient was 0.999 9.The detection limit for serum iodine was 1.2 μg/L (0.20 ml of serum was tested).Precision:the range of the intra assay coefficient of variation (CV) was 0.2%-1.2% (n =6) and the range of the inter assay CV was 0.4%-1.9% (n =3) when measuring 5 serum samples.Accuracy:The average recovery was 97.9% with a range of 93.9%-103.8% when measuring 8 serum samples.No significant difference was found between the results of the 35 serum samples determined by the standard method and the new method (t =0.178,P > 0.05).Conclusions Re performs better than Te in determination of iodine as an internal standard element.The reagent system screened for determination of iodine in serum does not require matrix matching.This method has good standard curve linearity,high sensitivity,good precision,accuracy and anti-interference ability,and is easy to be used and quickly to be analyzed of the test results,which is suitable for widely application in determining serum iodine.

ABSTRACT

Objective To evaluate the value of histogram parameters of dynamic contrast-enhanced MRI (DCE-MRI) in the diagnosis of cellular leiomyoma. Methods Seventy one patients with leiomyomas confirmed by pathology who underwent preoperative DCE-MRI were enrolled in this retrospective study. Quantitative perfusion histogram parameters (including median, mean, skewness, kurtosis, energy, entropy) were measured for each patient. Leiomyoma was divided into degeneration, ordinary and cellular types according to pathology. One-way analysis of variance and Least significant difference were used to compare the differences among the parameters of the three groups which were normal distribution and equal variances, while Kruskal-Walls test and Mann-Whitney U test were used to compare the parameters that did not conform to normal distribution or variance. ROC curves were drawn to evaluate the diagnostic efficiency of different parameters. Results Among the three groups,the values of Ktrans(median, mean, kurtosis, energy, entropy), Kep(median, mean, skewness, kurtosis, entropy), Ve(median, mean, skewness, kurtosis) and Vp(median, mean, skewness, kurtosis, energy, entropy) had statistical difference (all P<0.05). The values of Ktrans, Kep, Vp (median, mean) and Ktrans (entropy) of the cellular group were higher than those of the degeneration and ordinary groups. While the values of Ktrans (kurtosis, energy) and Kep (skewness) of the cellular group were lower than the other two groups;then the value of Kep (kurtosis) of the cellular group was lower than the ordinary group and the value of Kep (entropy) of the cellular group was higher than the ordinary group;and the values of Ve (median, mean, kurtosis) and Vp (entropy) of the cellular group were higher than those of the ordinary group. The values of Ve (skewness) and Vp (skewness, kurtosis and energy) of the cellular group were lower than those of degeneration group. ROC curves found that when Ktrans(median)was 0.994/min, its sensitivity was 100.0%, the specificity was 73.8%, and the area under ROC was 0.905;when Ktrans(mean) was 1.170/min, its sensitivity was 90.0%, the specificity was 85.2%, the area under ROC was 0.921. And the areas under ROC of Ktrans (kurtosis, energy, entropy), Kep (median, mean, skewness, kurtosis, entropy), Ve (median, mean, skewness), Vp (median, mean, skewness) were also high (ranging from 0.711 to 0.872). Conclusion Histogram analysis of DCE-MRI quantitative perfusion is applicable for identification of cellular leiomyoma.

ABSTRACT

Objective To investigate the influence and clinical significance of desensitization therapy on pe-ripheral blood type Ⅱ innate lymphocytes(ILC2s)in children patients with bronchial asthma.Methods 30 children patients with bronchial asthma in this hospital from January 2014 to December 2015 were selected and randomly divided into the simple medication treatment group(n=15)and medication combined desensitization treatment group(n=15),meanwhile 15 healthy children undergoing physical examination were selected as the control group.The flow cytometry and enzyme-linked immunosorbent assay were used to detect peripheral blood ILC2 and IL-13,IL-15,IL-33 expression levels before and after treatment.Results The peripheral blood ILC2s level in the simple medication treatment group and medication combined desensitization treatment group was significantly higher than that in the control group(P<0.05);the peripheral blood ILC2s level after 1-year treatment in the simple medication treatment was still higher than that in the control group(P<0.05);the pe-ripheral blood ILC2s level after 2-month treatment in the medication combined desensitization treatment group was decreased compared with before treatment(P<0.05),which had no statistical difference compared with the control group(P>0.05).The peripheral blood IL-5,IL-13 and IL-33 levels in the simple medication treat-ment group and medication combined desensitization treatment group were significantly higher than those in the control group(P<0.05).The peripheral blood IL-5,IL-13 and IL-33 levels after 1-year treatment in the simple medication treatment group were still higher than those in the control group(P<0.05);the peripheral

ABSTRACT

Objective@#To explore the effects and possible mechanism of rapamycin (RAPA) on apoptosis of CD4+CD25+ Tregs from the mouse severe aplastic anemia (SAA) model.@*Methods@#The BALB/c female SAA model mice were induced by interferon-gamma in combination with busulphan. The SAA model mice were intraperitoneal injection with RAPA at daily dose of 0.5 mg/kg for 5 days (the RAPA-treated group, n=15) in the SAA group (n=15) and the un-treated group (n=15) were control. Bone marrow hematopoiesis changes were observed by the patho-morphological examination of femurs. The mononuclear cells of the peripheral blood and spleen were subjected to assess the intracellular Foxp3 expression in CD4+CD25+ Tregs by flow cytometry (FCM). In addition, after being pured by immunomagnetic beads, the splenic CD4+CD25+ Tregs was subjected to assess apoptosis by FCM and the Akt and Stat3 phosphorylation by using of western blot.@*Results@#The patho-morphological examination of femurs showed normal marrow cell proliferation in un-treated group and hypocellularity in both SAA group and RAPA-treat group, with an increase in the number of fat cells. The bone marrow hematopoietic tissue ratio in RAPA-treat group was higher than SAA group [(9.75±1.83)% vs (7.00±2.00)%, Δx=2.15% (95%CI 0.15%-5.35%), P=0.037]. In the SAA group, FCM analysis showed down-expression of Foxp3 in CD4+CD25+ Tregs compared with the un-treated group. However, after treatment with RAPA, the expression of Foxp3 in CD4+CD25+ Tregs was increased (P<0.017). Compared with the un-treated group, increased CD4+CD25+ Tregs apoptosis [(19.84±1.39)% vs (29.85±2.72)%] with increased Akt phosphorylation accompanied by increased Stat3 phosphorylation was found in SAA group (P<0.05, respectively). On the contrary, RAPA-treated group exhibited CD4+ CD25+ Tregs with a reduction in apoptosis rate [(22.39±3.71)%], Akt phosphorylation and Stat3 phosphorylation compared with the SAA group (P<0.05, respectively).@*Conclusion@#These results indicate that RAPA may increase the expression of Foxp3 by down-regulation the levels of Akt and Stat3 phosphorylation and reduce apoptosis in splenic CD4+CD25+ Tregs from the mice model of SAA.
